Human Embryonic Stem (hES)

Human embryonic stem (hES) cell has a unique ability of differentiating into all cell types, which leads to the development of the whole organism. As the integrity of ES cells is crucial for the developing embryo, these cells have likely evolved a mechanism that will detect and respond to adverse stimuli. Indeed, hES cell has been shown to be highly sensitive to DNA damage, but the molecular mechanisms underlying this rapid death remain unclear. Caspases are critical mediators of apoptosis in the cells of mammals, and is considered a key protein that is responsible for controlling their activation is Bax, a Bcl-2 family proapoptotic member. While the main components of the apoptotic pathway have been known and identified, exactly how this pathway is functioning and regulated in various primary cells is still unclear. The study examined the apoptotic pathway in the hES cells and also reported a unique mechanism of hES cells that can help them to undergo apoptosis in response to the genotoxic damage. To visualize GFP-tagged Bax, the 3-day colonies of hES cells were transfected with 2 mg of hBaxC3-EGFP from Addgene with FuGENE HD transfection reagent from Roche. Transfection is the process of by which nucleic acids is introduce to the eukaryotic cells by methods that are nonviral. With the help of some various methods such as chemical, physical, lipid methods, this technology of gene transfer technology is a very powerful tool to investigate the gene function and the expression of protein of a cell. Assay-based reporter technology, and with the availability of transfection reagents will actually give and provides the foundation necessary to study sequences of mammalian promoter and enhancer, the trans-acting proteins such as transcription factors, processing of mRNA, the interactions of different proteins, recombination, and translation events. In this study, they describe the striking feature of the healthy undifferentiated hES cells, which maintain Bax in its preactivated state at the Golgi that is in contrast to other cell types. The results also highlight the fact that the apoptotic machinery undergoes a dynamic change even if its an early stages of differentiation.

In order to address this issue we have to take into consideration the historical background that has led to this notion. I am going to concentrate on one of the people that made the most important steps on the concept of production relating their work to its current form. I’m going to address the work of F. W. Taylor that was the theorist that put together what Henry Ford, the second persona, put into action through the assembly line. Taylor’s aim was to optimise the production of a product by focusing on two main aspects. Taylor’s theoretical contributions were justified my his belief that the one best ethod and best implementation can only be discovered or developed through scientific study and analysis†(Taylor,1964, 131) but his analysis failed to consider the implications that his methods have brought upon the workforce; even though they may have renovated the industrial world its side effect was the creation of a new generation of deskilled and demotivated workers. And as we observe this is not an issue of the past since all the assembly lines still work within the framework that Taylor f irst implemented, from a highly technological product to the simplest household item. This paragraph is going to focus on the problems that appear by the deskilled labour force and how this affects its employability. Deskilling as it’s defined by the Oxford Dictionary is â€Å"†¦the process by which skilled labor within an industry or economy is eliminated by the introduction of technologies operated by semiskilled or unskilled workers. † This elimination raises a number of issues, firstly is the issue of the working satisfaction of the labor force. By stripping the creativity out of the labor force’s job you are creating a level of dissatisfaction. This dissatisfaction is due to the fact that the reward that the workers get by the creativeness that they used to assert is now lost; even if in most cases it has been replaced by a higher payroll this didn’t seem to cover the damage. As Noon and Blyton (1997, 147) put it â€Å"The dehumanising effects can often be seen in the glazed expressions of the young people who serve. Thus you have a new generation of deskilled workers that lacks the confidence to interact with their working place or any working place, a worker that’s alienated who is not only less able to keep up with the increasingly demanding working environment but is less able to mobilize in any other work place. The other issue that arises from this deskilling that exists in the secondary sector is that for a deskilled worker it’s easier to become redundant. This is the case since the semiskilled workers may have gone through a sophisticated and elaborated training but their training only covered the absolutely necessary aspects of their specific task. This may be considered as a form of upskilling but in its essence is just another component making the worker more dependable to his current job in an epoch that the job market is becoming more and more contestable. The rise in the employments market contestability is largely dependable to the role of globalisation and up to a certain extend has played a main role to the instability that is currently present. Financial Times (Stern, 2012) defines globalisation as â€Å"†¦the integration of economies, industries, markets, cultures and policy-making around the world. † Therefore one cannot argue that it has contracted the employment market as well as any other market. This contraction is a result of the â€Å"widening deepening and speeding up of worldwide interconnectedness† (Baldry, 2007, 40) providing chances for people to compete in any market from anywhere in the world. That’s not an issue to the well rounded and highly trained workers, but it raises a number of concerns for the semi-skilled workers that add up to the overwhelming majority of the working population. They are faced with a crisis that not only have not caused but are also unable to confront. They have to compete against people from countries that are less paid and better trained causing the unemployment figures to rise dramatically as the IMF(Slaughter and Swagel, 1997) states here â€Å"there is no doubt that globalisation has coincided with higher unemployment among the less skilled and with widening income inequality. Other than that there are cases of mass shifts in the markets where companies assign projects to foreign countries that were previously based at home. We observe that for example in the telecommunication industry where the telephone helpdesks of British companies get assigned to Indian stated firms that offer a better deal due to the lower wages provided to the Indian workforce; making this abstract, as it is generally perceived, mobilisation of the industry more rigid and more challenging. Coming to an end I can’t avoid emphasizing on the statements vitality and the public concern that this raises. A statement that has never been more accurate than now; and as we observe notions such as Globalisation and Division of labour rooting and becoming embedded to our culture and our society this issue will not only rise but become untreatable. Resistance to change by employees, this problem shows when Dick as the plant manager asked the worker why he was using the saw when the material could easily be bent fifted into the barrels, resulting in saving time and equipment. After all the power saw are removed from the scalp area in order to adjust new method of bending rather than cutting. A few day later Dick find out that workmen using hand shears to cut each strip to cut. 2. Process of Production Problems. In his new position as Plant Manager at Modrow, he found that construction was slower in completion than originally planned; equipment arrived before the building was finished, employees were upset about the extent of change expected in their work routines with the installation of additional machinery, and in general morale was at low ebb. 3. Culture Change Problem; there is the difference between American and British philosophy and performance within the plant. Here, he also found that his power and authority were less than those of his superiors, so he constantly found himself in the position of having to soft pedal or withhold suggestions that he would have liked to make, or innovations that he would have liked to introduce. After a frustrating year and a half, Dick was suddenly made plant manager of an old British company which had just been purchased by Tri-American. He left his first English assignments with mixed and moved from London to Birmingham. In Birmingham, he become a plant manager and operated much as he had in his troubleshooting job for the first couple of years of his change from sales to administration. He did some changes in Birmingham. Training and reduction programs were instituted for all supervisors and managers who survive the initial purge. He tried to focus/ control on relationship between production and the needs from sales by simplifying and redesign in order to increase profit. Later the company showed a small profit for the first time in many years. For this successfulness, then he requested transferred back to United States. 4. Cost Reduction Problem, by the end of his first year at Modrow, the building program had been completed, although behind schedule, the new equipment had been installed, and some revamping of cost procedures had been incorporated. But, the financial picture at this time showed as a loss. Later, all managers of the various divisions had worked closely with their supervisors and accountants in planning the budget for the following year. 5. Human Relations/ Interaction Problems When he worked through his first year as plant manager, Dick developed the habit of strolling around the organization. He was apt to leave his office and appear anywhere on the plant floor, in the design office and at the plant cafeteria rather than in the executive dining room, or wherever there was activity concerned with Modrow. At first his presence disturbed those who noted him coming and going, but later, the workers came to accept his presence and continue their usual activities. Supervisors, managers, and foreman, however, did not feel as comfortable when they saw him in the area. 6. Physician-assisted suicide refers to the physician acting indirectly in the death of the patient -- providing the means for death. The ethics of PAS is a continually debated topic. The range of arguments in support and opposition of PAS are vast. Justice, compassion, the moral irrelevance of the difference between killing and letting die, individual liberty are many arguments for PAS. The distinction between killing and letting die, sanctity of life, "do no harm" principle of medicine, and the potential for abuse are some of the arguments in favor of making PAS illegal. However, self-determination, and ultimately respect for autonomy are relied on heavily as principle arguments in the PAS issue. Daniel Callahan, author of When Self-Determination Runs Amok, is against any social policy that would allow for PAS to be practiced. Callahan believes that the argument for PAS does not have a firm foundation, because self-determination and mercy, the two principles that are in support of PAS, may become separated (711). If mercy is seen as a core element in support of PAS, why restrict PAS only to those who can ask for it -- don't the unconscious or incompetent deserve mercy also? Callahan is in opposition to the belief that the essence of human dignity is the notion that a person should be free to choose how and when they want to die. Callahan questions the absolute nature of autonomy and self-determination and the extent to which these values can be applied. Self-determination by definition entails human freedom of action and respect for persons but those in support of PAS want it to be restricted to those who are terminally ill. Human suffering and an individual‘s outlook on the quality of their life, are, in Callahan’s opinion, subjective and there is no one standard to compare individual suffering. If we just focus on autonomy/self-determination, why restrict PAS only to those who are terminally ill and competent? Are the incompetent less deserving of relief from suffering than the those competent individuals? If physician-assisted suicide is legally permitted yet restricted to the terminally ill adult with full decision-making capacity, it will certainly raise legal concerns about discrimination. PAS will probably broaden to include incompetent, non-consenting, and non–terminally ill persons. The final extreme of the slippery slope argument is that PAS will be abused, run amok and ultimately become involuntary euthanasia. Teaching Preschool Children Reading

Phonological awareness is an important aspect in the fundamentals of reading. It is the first step in literacy. Children can learn phonological awareness in a variety of ways. Rhyming, sentence and word recognition, sound knowledge, phonemes, letter identification, spelling, and games which require active listening are a few of the techniques used by teachers in the classroom. All of these methods help in a child’s reading and speech fluency.All children need to learn how to read and write in order to survive in today’s society. There have been many different methods used in order to teach phonemic awareness to preschool children. Phonemic awareness is the capability of an individual to differentiate, identify and manipulate specific sounds. An example would be a child combining and blending the sounds in the word cat. Many people believe that phonics and phonemic awareness are the same thing. However, phonic awareness is recognizing that words come from sounds with in turn make up a language. Phonics is knowing that sounds are composed of letters which compose a writing style.In order to teach a child phonic awareness and to read, a teacher must comprehend the procedure that the brain formulates in order to understand the printed data. The brain undergoes three functions which facilitate understanding. First, there is information retention which has to deal with spurs of the environment. Second, language articulation is where a child uses prior knowledge in order to associate its meaning; and the third process is modeling and making connections with former information . Reading comprehension occurs in the frontal lobe of the brain. When this process takes place, it leads to an understanding and knowledge of what a person has seen and read.Phonological processing is a person’s ability to listen and  comprehend dialect as well as printed terms. Hence, it is to say that the child recognizes the sound each letter makes. Mechanically our brai n unconsciously processes verbal communication. Instantly a child gathers phonemes in order to pronounce the word and decomposes it to grasp the oral language. In contrast to speech, reading involves a child’s knowledge in the process of associating written words with the alphabet and in turn producing spoken words. Therefore, a child has to be of conscious mind in order to learn reading. A teacher must instruct a child in the phonological sequences of letters in order for the child to acquire phonological processing.There are many different techniques a teacher can utilize in order to build phonological awareness. One technique would be merging words and sounds collectively in order to construct new words. The words composed can be silly or factual. This technique facilitates the child’s use of phonemes, which was a study conducted by McCandlies, Cohen, and Dehaene in 2003 . Having a child combine letters and then separating them is another form of building phonologic al awareness. It will also help them in recognizing the letters of the alphabet. Examples of the activities mentioned above are having a child say the first sound in the word â€Å"rat† and then saying each letter sound individually. Blending would consist of telling a child say ‘at’ and then stating to them to put an ‘r’ in front and say the new word.These activities can been done with a variety of materials such as chalk, boards and markers. Remember preschool children are very visual. The more movement they utilize, the better they retain the information. Image is a phonic awareness 3 letter word game.Many people do not see imaginative play as a means of learning. However, it is during play that children are at their most influential developmental stage according to Vygotsky, a Russian psychologist. Since the children are imaging different situations, they are associating everyday activities and assigning meaning to them. During this time, individ uals and items can transform themselves into different things. For example, a child can become a cat, or a baby and a piece of construction paper can be converted into a notepad for taking orders in a restaurant. Through the children’s interactions and language, they gather and exchange knowledge, experiences,  and vocabulary which enhance their verbal communication through mutual exchange (Genishi and Dyson, 2009).Collaborative songs and games can also be a means to teach phonological awareness. Nursery rhymes can be utilized as a tool for phonological awareness. Rhyming activities such as asking a child which words rhyme or which ones do not rhyme will enhance their awareness. A teacher may also say a word and ask a child which word rhymes with it (Phillips, Clancy-Menchetti, Lonigan, 2008). Some examples would be: this old man, head shoulder knees and toes, one two buckle my shoe and Humpty Dumpty. Games that constitute phonological awareness do not have to come from a b ook. They can be invented from one’s everyday activities. Examples of these activities would be a trip to the grocery store or beauty salon.Children can be asked which items in the store begin with the same letter sound or which nail polishes have the same letters in them. If a child does not respond or say the correct answer, be sure to mirror the correct response and not state that they were wrong. The teacher should support the child through the learning process. The following illustration explains the steps of phonological awareness.Activities which implement pattern recognition are also efficient at phonological awareness. This is an excellent tool to utilize in the classroom due to the fact that children learn how to read by identifying patterns. When a child associates words from letter patterns and connects the sounds, this is called patterning. A teacher should replicate, rehearses, and observe the children when teaching pattern recognition in order to determine the child’s advancements in this area. Assessments should cover word subdivision, sound combinations, and sorting new data into patterns. Repetition is the key in the learning process.Preschool children have a short attention span; therefore, keep activities fun and exciting in order to facilitate the best learning environment. Not all children have the same literacy background and predisposition; they come from a variety of environments and cultural upbringings. Consequently, teachers must adapt to each individual child’s style of learning and pace. There will always be  variations in the learning process.Technology is a part of our everyday lives. Children have computers, iPhones, iPod’s, and tablets. Since technology surrounds children twenty -four hours of the day, seven days a week, parents need to take advantage of the situation and use the time wisely.
